As with 5.1, the route selection is SOR1, SOR2, and a blend of SOR3 & Remake.

Thanks, so if I select route o the 2nd game, all stages will be in the same order of the original game until the last stage and last boss of the 2nd game?? Or once I reach the last stage of the 2nd route/2nd game the game will automatically continue from the first stage of the 3rd route/3rd game??

No, it's always an 8 stage route. There are some branching paths you can take, but there's no marathon option, unless someone has made a mod for one. If I remember right, you can choose between the SOR1 & 2 route to switch on Stage 4, and the SOR2 Stage 6 jungle in Remake has a path that splits off for a hidden Stage 7. The big changes in 5.2 from 5.1 are the Waterfront scene, an updated Wasteland, and the Sunset jetski section has an island at the half-way point.
